POSITION SUMMARY
The individual will investigate the role of epigenetic regulators/chromatin factors in transcriptional regulation of endogenous retroviruses. This will constitute performing an siRNA-mediated knock-downs followed by qRT-PCR, bisulfite sequencing and Chromatin immunoprecipitation.
The second project to be carried out by the individual is a small molecule screen aimed at identifying inhibitors of SETDB1. The individual will perform the screen and follow up with experiments confirming positive hits employing qRT-PCR, cell cycle and proliferation analyses.
QUALIFICATIONS:
Must be proficient in mouse embryonic stem cell culture, siRNA and plasmid transfections, molecular cloning, qRT-PCR, flow cytometry and standard molecular biology techniques.
Must have extensive knowledge of the fields of endogenous retroviruses, mouse development and epigenetic regulation.
Basic understanding of genetics and molecular biological techniques is required. Background in the study of repetitive elements essential, >3 yrs postdoctoral experience preferred.
ORGANIZATIONAL STATUS
The individual reports to the principal investigator. Results are reviewed and additional experiments outlined on a weekly basis. Interacts with members of this and other laboratories in the Molecular Epigenetics Group.
CONSEQUENCE OF ERROR/JUDGEMENT
The analyses performed require strong background in molecular biology. Errors will result in delayed output of data and publications.
SUPERVISION RECEIVED
After initial training the incumbent is expected to perform the above experiments and analyses with only general supervision. Routinely discusses results and plans for additional analyses with principal investigator on a daily or bi-weekly basis.
SUPERVISION GIVEN
Individual performs molecular biology and teaches these techniques to other lab members.
WORKING CONDITIONS
Works in a laboratory with chemicals and biologicals. Radioisotopes, microbes, and recombinant DNA are routinely handled.
